SANOVO (NCT05009836) is a double-blind phase 3 trial in China testing first-line savolitinib plus osimertinib vs osimertinib alone in 326 patients with EGFR-mutant, MET-overexpressing NSCLC. At ESMO 2026 (LBA69), investigator-assessed PFS was 24.9 vs 13.8 months (ITT, HR 0.56). The first-line combination is investigational.
See the LBA69 Results →The primary endpoint, investigator-assessed PFS, was tested hierarchically: the MET IHC 3+ population first, then the ITT population (IHC 3+ and IHC 2+). Both were met at the data cutoff of July 31, 2026. In MET IHC 3+ (98 vs 98 patients; median follow-up 30.4 months), median PFS was 24.8 months (95% CI 19.4–27.6) with savolitinib plus osimertinib vs 11.1 months (8.3–16.6) with placebo plus osimertinib: HR 0.45 (0.31–0.66), P < 0.0001. In the ITT population (162 vs 164; median follow-up 38.6 months), median PFS was 24.9 months (22.0–27.6) vs 13.8 months (10.9–16.6): HR 0.56 (0.43–0.74), P < 0.0001. HUTCHMED’s release prints the ITT combination-arm CI as 22.1–27.6.
INV PFS: IHC 3+ HR 0.45 · ITT HR 0.56Independent review committee (IRC) assessment, a secondary endpoint, was consistent: MET IHC 3+ median PFS 22.2 months (19.4–NE) vs 13.7 months (8.3–19.4), HR 0.48 (0.32–0.72); ITT 27.9 months (22.1–35.8) vs 16.4 months (9.7–16.6), HR 0.53 (0.39–0.71) (median follow-up 33.1 months). These IRC figures are separate from the primary investigator-assessed results above; the discussant’s first-line comparison slide used the IRC medians (ITT 27.9 vs 16.4 months; IHC 3+ 22.2 vs 13.7 months).
IRC PFS: IHC 3+ HR 0.48 · ITT HR 0.53In the ITT population, with 49 (30.2%) vs 83 (50.6%) deaths and 40.5% maturity (median follow-up 38.7 months), median OS was 44.5 months (41.3–NE) vs 34.9 months (26.3–41.1), HR 0.48 (0.33–0.68). OS rates were 94.4% vs 81.0% at 12 months, 79.8% vs 65.4% at 24 months and 67.4% vs 47.4% at 36 months. No P value was reported for OS; the abstract describes a “clinically meaningful OS benefit.” Crossover between arms was not allowed. Subsequent anticancer therapy was given to 46.9% vs 59.1%, including a MET TKI in 1.2% vs 4.3%.
OS 44.5 vs 34.9 mo · HR 0.48ORR was 80.9% (74.0–86.6) vs 67.7% (59.9–74.8), odds ratio 2.00 (1.17–3.45); disease control 97.5% vs 88.4%. Median duration of response was 23.5 months (20.1–26.4) vs 15.2 months (12.4–18.0).

In China it is approved with osimertinib for EGFR-mutant non-squamous NSCLC with MET amplification after EGFR-TKI therapy (SACHI), for MET exon 14 skipping NSCLC, and conditionally for MET-amplified gastric or gastroesophageal junction adenocarcinoma after at least two prior systemic therapies. ✓ Osimertinib (TAGRISSO®, AstraZeneca) alone is FDA approved for first-line EGFR-mutant NSCLC. All patients were enrolled in China. The combination after osimertinib was tested separately in SAFFRON.

The ITT population included IHC 3+ (≥90% of tumor cells with strong staining) and IHC 2+ (H-score ≥220). Riyaz Shah, MD, called it “3+ or 2+ (interesting definition)”. In the investigator-assessed subgroup analysis the IHC 2+ HR was 0.85 (0.57–1.27), and the discussant, Rebecca Heist, MD, asked “Is there benefit in the 2+ population?” and “What is the right MET definition in the 1L population?” Ryan Gentzler, MD, noted “Questions remain around how to test for MET positivity (IHC, amplification, cutoffs)”.

SANOVO asked whether adding savolitinib, an oral, selective MET tyrosine kinase inhibitor, to osimertinib improves progression-free survival as first-line treatment for EGFR-mutant advanced NSCLC with MET overexpression, a group with shorter benefit from EGFR-TKI monotherapy. The trial is run by HUTCHMED (CT.gov sponsor) and funded by HUTCHMED and AstraZeneca.
Randomized 1:1, double-blind, placebo-controlled phase 3 at 74 centers in China (NCT05009836). 326 randomized (162 vs 164), enrollment August 2022 to August 2025. Stratified by EGFR mutation type, MET status (IHC 3+ vs 2+) and brain metastases. Crossover not allowed.
Untreated unresectable or metastatic NSCLC; EGFR exon 19 deletion or L858R; centrally confirmed MET overexpression by IHC (D1C2, Amoy Diagnostics): IHC 3+ (≥90% of tumor cells with strong staining) or IHC 2+ (H-score ≥220 and <90% strong staining); ECOG PS 0–1. 196 patients (98 per arm) were IHC 3+; brain metastases at baseline 38.9% vs 40.2%.
Savolitinib 300 mg or 200 mg twice daily (by body weight) or matching placebo, plus osimertinib 80 mg once daily, until progression or intolerable toxicity.
Primary: investigator-assessed PFS (RECIST 1.1), tested hierarchically in MET IHC 3+ then ITT. Secondary: IRC-assessed PFS; ORR, DoR and DCR by investigator and IRC; OS; safety.
Presenter: Yi-Long Wu, MD (Guangdong Provincial People’s Hospital), lead principal investigator. Invited discussant (LBA69 and 2615O): Rebecca S. Heist, MD (Massachusetts General Hospital).
Data cutoff July 31, 2026. Median follow-up for PFS 30.4 months (IHC 3+) and 38.6 months (ITT, investigator); for OS 38.7 months, OS maturity 40.5%.
SANOVO (NCT05009836) is a randomized, double-blind phase 3 trial at 74 centers in China, run by HUTCHMED, comparing savolitinib (a MET inhibitor) plus osimertinib with placebo plus osimertinib as first-line treatment in 326 patients with EGFR-mutant (exon 19 deletion or L858R) advanced NSCLC and centrally confirmed MET overexpression (IHC 3+ or IHC 2+). Design: Trial Design.
At the data cutoff of July 31, 2026, investigator-assessed PFS, the primary endpoint, improved in both hierarchically tested populations: MET IHC 3+ 24.8 vs 11.1 months, HR 0.45 (95% CI 0.31–0.66), and ITT 24.9 vs 13.8 months, HR 0.56 (0.43–0.74), both P < 0.0001. Median OS, a secondary endpoint, was 44.5 vs 34.9 months, HR 0.48 (0.33–0.68), at 40.5% maturity; no OS P value was reported. Figures: At a Glance.
SANOVO tested savolitinib plus osimertinib as first-line treatment in untreated, MET-overexpressing EGFR-mutant NSCLC, against osimertinib alone. SAFFRON tested the same combination after progression on osimertinib, against platinum-pemetrexed, in patients with MET overexpression or amplification. Different setting, comparator and biomarker definition. See SAFFRON.
No. The first-line combination is investigational everywhere; HUTCHMED plans a supplemental NDA in China based on SANOVO. Savolitinib is not FDA approved. In China it is approved with osimertinib for EGFR-mutant non-squamous NSCLC with MET amplification after EGFR-TKI therapy (SACHI), and alone for MET exon 14 skipping NSCLC. Osimertinib alone is FDA approved for first-line EGFR-mutant NSCLC.
Grade ≥3 treatment-emergent adverse events occurred in 77.2% vs 38.4%, serious events in 54.9% vs 28.7%, and events leading to discontinuation in 14.8% vs 4.3%; fatal events (excluding progression) were 4.3% vs 4.8%. Median exposure was 22.1 vs 12.9 months. The most common events with the combination were peripheral edema (65%), hypoalbuminemia (63%), decreased platelet count (51%) and anemia (49%).
